The immunotherapy of cancer has been an elusive dream in oncology, but Dr. Calabrese will illustrate the recent revolution of such evolving therapies by the therapeutic targeting of checkpoints. The results represent one of the major advances in applied immunology over the past decade or more. Early targets have included CTLA-4 and PD-1 with multiple approved therapies, and clinical trials are being initiated with these and other regulatory proteins. Insights into regulating checkpoints are now incorporated into the earlier stages of treatment of refractory opportunistic infections and autoimmune diseases.
